Phase I: Antigen Selection

We can use a range of macromolecules as antigens, including:

Phase II: Fusion, Expansion, and Screening

After immunization, spleen B cells are fused with myeloma cells to form hybridomas, which are cultured under selective conditions. Positive clones producing antigen-specific antibodies are identified through high-throughput screening (e.g., ELISA or FACS) and expanded. Top candidates are selected for subcloning to ensure monoclonality and stable antibody expression. This phase includes a 7-step process which typically takes 8–10 weeks to complete.
